Culture_conditions 80% RPMI 1640 + 20% FBS + (10 ng/ml mIL-3) split saturated culture 1:2 every 2-3 days; seed out at ca. 0.5 x 106 cells/ml; maintain at ca. 0.5-1.0 x 106 cells/ml; cells are very difficult to culture, mIL-3 supports growth of the cells, cells grow best in 12- or 24-well-plates; after thawing, for the first 7-10 days there is virtually no cell growth or cells grow very slowly, viability may be initially very low, not exceeding 80-85% at 37C with 5% CO2 maximum density at about 1-1.2 x 106 cells/ml
